I would choose the C43 (of course I am a little biased though). I think that it is a great mix of comfort, sound, styling, and speed.
There are more than enough mods for it as well. You can actually swap engines with the old E55, ML55 engine if you wish. Also, Kleemann et. al. offer a lot of options.
The going rate seems about right for what you are being quoted.
Mainly, it is about finding a car you are happy with. Try them all and see what you think. If, however, you are looking for a fast car and something with good potential to modify, then I would suggest the C43.

Depends on whether you are performance minded and would "use" a hi-performance car... C43 is w/o a doubt the leader in that list. It also has a fold-down seat I don't think the E320 has. Though if you have a family, the E has more legroom for those in the rear.
As I'm one with a 5.4L transplant, ya that's really not that hard (probably more $ then a supercharger tho). I don't think Kleemann is selling superchargers for the 6-cylinders anymore, as there was very little demand.
Dunno about that BMW; I'd go for a 540 if I was going for a 5-series.tho.
The C43 is the "bargan" in that list IMO.

The transmission failure seems to be a gamble. Some cars have the problem, some don't. You can look in the "problems" area for more detail. I have written about it there.
Mine, however, has never had the problem. I am at 45K miles as of now.
I also have no problem with the speakers. I heard a buzzing noise at some point, but found it to be due to some loose screws in the rear deck.
